报告人简介

柯子斌1992年毕业于香港大学,获得生物学士;1997年在香港大学获得分子生物学博士学位。现为香港理工大学应用生物与化学系研究助理教授;應用生物及生物科技課程副主任。主要的研究领域为肝细胞癌的发生、发展、转移的分子机制;筛选中药材中天然的小分子药物;蛋白在细胞核与细胞质内的穿梭机制。此间发表SCI论文四十多篇,包括在Hepatology Cancer Research杂志上。应邀综述3篇。所有论文被引用次数达到3800次。此间獲得政府研究經費大約二千萬。

CCCTC-binding factor (CTCF) is a highly conserved nuclear factor involved in the regulation of transcription, insulation, and chromatin architecture. Mutations of CTCF-binding sites were shown to be associated with carcinogenesis of various cancer. We made a pivotal discovery that CTCF expression is low in liver but frequently upregulated in hepatocellular carcinoma (HCC) and associated with poor prognosis. We elucidate the functional role of CTCF in HCC using cells andin vivomouse models. Cell and xenograft analysis showed that depletion of CTCF in HCC cells reduced cell growth, migration and invasion, whereas overexpression of it promotes tumor growth, cell mobility and invasiveness. I will discuss the putative functions of CTCF in HCC carcinogenesis using our latest findings from transcriptomic profiling and ChIP-seq analysis. Our data suggested that targeting CTCF could be further explored as a potential HCC therapeutic strategy.
